Photoimmunology · An introduction

How light rewires your skin's immunity.

Photoimmunology explores the interplay between solar ultraviolet light and the skin's immune system - how light shapes cutaneous immunity, for harm and for healing. Scroll to go deeper.

02 · The first hit

Damage and a switch

UV radiation strikes two epidermal targets: it damages cellular DNA - forming cyclobutane pyrimidine dimers - and converts trans-urocanic acid in the stratum corneum into cis-UCA, which initiates systemic immune suppression.

03 · A change of orders

Tolerance, not alarm

Phenotypically altered Langerhans cells migrate to draining lymph nodes. Rather than activating defense, these tolerogenic dendritic cells prime naive T-cells into antigen-specific Regulatory T-cells (Tregs).

04 · The cascade

Stress and signals

UV triggers oxidative stress - reactive oxygen and nitric oxide species. Activated skin nerves release neuropeptides like CGRP, driving mast cell degranulation and a cascade release of immunosuppressive IL-10.

05 · Silent cleanup

Clearing the fallen

Severely damaged sunburn cells undergo programmed apoptosis. Immature dendritic cells quietly clear these apoptotic bodies through efferocytosis, reinforcing immune tolerance rather than inflammatory alarm.

06 · The dual edge

Harm and healing

The same immune modulation cuts both ways: uncontrolled UV drives photocarcinogenesis by suppressing tumor clearance, while targeted phototherapy harnesses controlled immunosuppression to resolve inflammatory skin disorders.

The takeaway

Light is both a threat and a therapy.

Photoimmunology is what lets us limit UV's harm to immune surveillance - and turn controlled light into medicine for inflammatory skin disease.
